NovaSyn TG amino Resin (130 µm)

NovaSyn® TG Amino Resin (130 µm): Macroporous Resin for Fmoc-Based Peptide Synthesis

NovaSyn® TG Amino Resin is a macroporous polyethylene glycol (PEG)-grafted polystyrene resin functionalized with primary amino groups. Designed specifically for Fmoc solid-phase peptide synthesis (SPPS), this resin features uniform spherical beads (~130 µm) and excellent swelling characteristics in both polar and nonpolar solvents. The PEG grafting dramatically improves solvation and reagent accessibility, making it ideal for synthesizing long, hydrophobic, or aggregation-prone peptides.

The free amino groups on the resin enable custom first amino acid attachment, giving chemists flexibility to design C-terminal carboxylic acid peptides tailored to their synthesis goals.

Specifications

  • Matrix: PEG-grafted polystyrene (NovaSyn® TG)

  • Functional Group: Primary amine (–NH₂)

  • Bead Size: ~130 µm

  • Loading: 0.2–0.4 mmol/g (typical)

  • Swelling: Excellent in DMF, DCM, NMP, MeOH

  • Compatible Chemistry: Fmoc/tBu-based SPPS

Key Features

  • PEG-Grafted Macroporous Structure: Enhances reagent diffusion and swelling

  • Customizable: Free amino groups allow attachment of any first residue or linker

  • High Solvent Compatibility: Performs well in both polar and nonpolar solvents

  • Ideal for Difficult Sequences: Long, hydrophobic, or aggregation-prone peptides

  • Fmoc-Compatible: Fully suited for modern SPPS workflows

Applications

  • Custom C-terminal acid peptide synthesis

  • Attachment of custom linkers or functional handles

  • Synthesis of challenging or long peptide sequences

  • Convergent synthesis strategies with fragment condensation

  • Peptide–drug conjugates and modified peptide systems

Attachment Example (Workflow)

  1. Swelling: Resin is swollen in DMF or DCM

  2. First amino acid coupling: Activated with DIC/HOBt, PyBOP, or similar reagents

  3. Capping (optional): Unreacted amino groups can be capped with acetic anhydride

  4. SPPS cycles: Standard Fmoc deprotection and elongation steps follow

  5. Cleavage: Based on the nature of the attached linker

Handling & Storage

  • Store dry in a sealed container away from moisture and light

  • Swell in DMF, DCM, NMP, or other compatible solvents prior to use

  • Stable under standard SPPS conditions

  • Avoid prolonged exposure to strong acids or bases unless intended for cleavage

Comparison with Other Amino Resins

Resin Matrix Type Loading Best Use
NovaSyn TG Amino PEG-grafted PS ~0.2–0.4 mmol/g Long/hydrophobic peptides, difficult sequences
Aminomethyl PS Resin Polystyrene ~1.0 mmol/g General SPPS, custom C-terminal acids
Tentagel® NH₂ Resin PEG-PS (swelling in aqueous media) ~0.2–0.3 mmol/g Peptides in water-compatible systems

Conclusion

NovaSyn® TG Amino Resin (130 µm) offers excellent performance for custom and advanced peptide synthesis, particularly where peptide solubility and chain accessibility are concerns. Its macroporous PEG-PS structure and free amino functionality make it a flexible and high-efficiency choice for modern Fmoc-SPPS.
