4-Fmoc-hydrazinobenzoyl AM NovaGel

4-Fmoc-Hydrazinobenzoyl AM NovaGel™ Resin

Specialized Resin for Hydrazide Peptide Synthesis (C-Terminal Hydrazine Derivatives)

4-Fmoc-hydrazinobenzoyl AM NovaGel™ Resin is a pre-functionalized polyethylene glycol (PEG)-grafted polystyrene resin designed for the solid-phase synthesis of peptide hydrazides—valuable intermediates for native chemical ligation (NCL) and other chemoselective ligation strategies. The resin incorporates a 4-Fmoc-protected hydrazinobenzoyl linker, which upon deprotection, generates a hydrazine-functional handle at the C-terminus of the growing peptide.

Built on NovaGel, a highly swollen PEG-polystyrene matrix, this resin offers excellent solvation, reagent diffusion, and is ideal for difficult sequences, long peptides, or sensitive fragments. It is fully compatible with Fmoc/tBu-based SPPS.

Key Features

Applications

Typical Workflow

  1. Swelling: Resin is swollen in DMF or NMP

  2. Fmoc Deprotection: Removal with 20% piperidine in DMF liberates the hydrazine group

  3. Peptide Elongation: Standard Fmoc SPPS proceeds using hydrazine handle as anchor

  4. Cleavage: Global cleavage with TFA provides peptide–NHNH₂ hydrazide product

  5. Post-synthesis modification: Hydrazides can be converted to thioesters or aldehydes

Cleavage Protocol

Reagent Conditions Result
TFA:TIS:H₂O (95:2.5:2.5) 1–2 hours at room temperature Fully deprotected peptide hydrazide

Storage & Handling

  • Store in a dry container under inert atmosphere

  • Protect from light, air, and moisture

  • Swell in DMF or DCM prior to synthesis

  • Use standard Fmoc deprotection and SPPS reagents
